CMV infection can be diagnosed\nvia urine specimens collected within 2–3 weeks after birth. We studied the epidemiology and treatment of CMV infection as well as\nanti-CMV drugs and vaccines. First, we developed a rapid and convenient urine collection method and CMV detection assay using\nfilter paper inserted into a diaper. Our study group, supported by funding from MHLW, screened 23,000 newborns using the CMV\nassay and found that the frequency of congenital CMV infection was 1 in 300 newborns, which is higher than any metabolic genetic\ndefects. Around 30% of the infected infants exhibited clinical abnormalities at birth. Epidemiological analyses indicated that childto-\npregnant mother was the main transmission route. Second, based on the protocol defined by the study group, some symptomatic\npatients were treated with anti-CMV drugs, and viral loads in these patients were analyzed. We found that treatment lasting >6 weeks\nwas sometimes required and that viral loads in plasma specimens correlated with clinical manifestations. Because side effects and the\ndevelopment of resistance limit the use of available anti-CMV drugs and because there are only a few drugs in clinical trials, there is a\nneed to develop new anti-CMV drugs. We established a CMV reporter cell line, screened 9,600 random compounds, and identified\nsome compounds with anti-CMV activity. One of these compounds, DPPC, inhibited an early step of infection and was effective in\nan animal model. Third, in the congenital CMV infection model of guinea pigs, we found that the current vaccine strategy based on\nglycoprotein B cannot block cell-to-cell viral transmission in the placenta.
